Tissue Culture Deflasking & Acclimation Guide 🌱

What is Tissue Culture?

Tissue culture consists of taking a portion of a plant (e.g. the apex, a leaf or segment thereof, stem segment, meristem, embryo, node, seed, anther, etc.) and placing it in a sterile nutrient media (usually gelled, semi-solid) where one or many plants will regenerate.

Thank you for your purchase! These are the steps we personally use, with an almost 100% success rate when acclimating our tissue cultures.

1. Let your TC rest (48–72 hours)

Before opening the bag or container, let your tissue culture adjust to your growing environment for 48–72 hours. Simply place it where it will be growing over the next few months.

2. Prepare your workspace

Sterilize your work area and gather:

  • Distilled water

  • Tweezers

  • Soft toothbrush or paintbrush

  • Paper towels

  • Alcohol

  • Fungicide

  • Rooting hormone

  • Transplant vitamin

  • Your preferred substrate

  • A humidity dome/container (100% humidity for the first 6 weeks)

3. Remove all the agar

Using sterilized tweezers, gently remove the plant and rinse the roots thoroughly in distilled water. Use a soft brush to remove as much agar as possible, as leftover agar may encourage rot.

Don’t worry if a few leaves or roots fall off—this is completely normal. Remove any dead tissue before planting.

4. Fungicide dip

Soak the plant in a diluted fungicide solution for no longer than 10 minutes, following the manufacturer’s instructions. Rinse again with distilled water afterward.

5. Vitamin soak

Soak the plant in distilled water mixed with transplant vitamin for 10–15 minutes (up to 30 minutes if desired).

6. Air dry

Let the plant rest on a paper towel for 5–10 minutes to remove excess moisture. Don’t allow the roots to dry out completely.

7. Potting

We recommend:

  • Tree fern fiber

  • AAA sphagnum moss (pre-soak at least 3 hours with rooting hormone)

  • Coco/perlite mix

  • High quality propagation plugs

Pot into a small container, moisten the substrate (not soaking wet), and maintain 100% humidity for the first 6 weeks.

Provide bright indirect light or grow lights for 12–16 hours daily. (We strongly recommend buying a cheap light meter from Amazon, aiming for 500-800 Foot Candles (FC) is the sweet spot for growing healthy & vigorous TC’s)

8. Acclimation

New roots should begin forming within 2–3 weeks. Even if you see roots, keep humidity high.

After 6 weeks, gradually reduce humidity by opening the container for a few hours each day over the course of a week.

Use fertilizer at ½ strength for the first 4 months, then care for your plant as you would any established plant.

DoseX (Rooting Hormone): 1 ml per gallon of water / .25 ml per liter.

ThriveX (Organic Fertilizer/Transplant Vitamin): 4 ml per gallon of water / 1 ml per liter.

FungiX (Fungicide): 5 ml per gallon of water / 1.25 ml per liter.

Each drop bottle contains 5 ml of each solution.
